abstract | A method of connecting reaction components to load sensors, comprising (a) providing a solid support comprising numerous load sensors, wherein each of the load sensors is a field-effect transistor and has an ability to connect to numerous reaction components; (b) providing a fluid comprising numerous reaction components of a particular type; (c) bringing the solid support into contact with the fluid under the conditions in which (i) the numerous reaction components of the particular type are in fluid communication with the numerous load sensors, (ii) in the fluid there are a greater number of reaction components of the particular type than the number of charge sensors on the solid support; and (iii) the reaction components of the particular type are connected from the fluid to the load sensors, thus forming modified load sensors that are connected to the multiple reaction components of the particular type of the fluid; and (d) removing, degrading or inactivating one or more of the reaction compounds of the particular type of each of the modified load sensors to leave only one of the reaction components of the particular type connected to at least one of the sensors of modified fillers wherein one of: (iv) removing involves cleaving cleavable anchors to which the reaction components are connected; or (v) degrading involves attaching a protecting moiety to the only one of the reaction components, and then degrading any of the unprotected reaction components; or (vi) inactivating involves linking a nucleotide analog chemically or photochemically to one or more active sites of the polymerase, and then applying a chemical or photochemical treatment to render the one or more of the reaction components inactive; or (vii) the removal, degradation, or inactivation of excess reactive components from the load sensors is monitored to determine the extent of removal, degradation, or inactivation; or (viii) the removal, degradation or inactivation is monitored with a different sensor to distinguish the presence of reaction components on the surface of the reaction components. |
