A polynucleotide comprising a transposon comprising inverted repeats of the ends of a piggyBac-like transposon flanking a heterologous polynucleotide not naturally flanked by inverted end repeats, inverted repeats are flanked on their outer sides by copies of a target sequence, wherein the transposon can be transposed by a transposase identical to SEQ ID NO: 45 fused to a heterologous nuclear localization signal (NLS) wherein SEQ ID NO: 124 provides the sequence of each inverted end repeat, the sequence is in inverted orientations at the two ends of the transposon. |
