abstract Procedure to increase the yield of IgG in a purification process of a production intermediate, comprising (1) fractionation with octanoic acid, (2) incubation at low pH, (3) pH shift and deep filtration, (4) chromatography of anion exchange, (5) virus filtration and concentration by ultra / diafiltration, in which step (3) of pH shift and deep filtration is modified to comprise (a) supply of an acid solution comprising IgG, other immunoglobulins and / or other protein contaminants with a pH between 3.5 to 5.2 and a total protein concentration of at least 10g / I; (b) adjusting the solution to a pH of 5.2 to 6.2 while maintaining a conductivity below 1.5 mS / cm; (c) incubation of the solution for at least 15 minutes; and (d) removal of any precipitate in which the pH adjustment in step (b) is accomplished by adding at least one multi-hydroxy amine compound with or without a carboxyl group.
