abstract The invention comprises 2-amino-(mono- or di-substituted)-ethyl guanidines having the formula <FORM:0972888/C1/1> wherein R1 and R2 are different and each is either a hydrogen atom or an alkyl, cycloalkyl or aralkyl group with 1 to 7 carbon atoms, and their addition salts with organic or inorganic acids. The compounds are prepared by condensing the corresponding N1-mono- or N1,N1-di-substituted ethylene diamine dissolved in water or an organic solvent, with cyanamide, its salts, or a compound having the formula <FORM:0972888/C1/2> wherein X is an alkylmercapto, alkyloxy, or pyrazolyl group. Organic solvents which may be employed are alcohols, ethers, ketones and C1-4 aliphatic acids. The reaction may be carried out at room temperature but is preferably carried out at a temperature up to the boiling point of the reaction solvent for 3 to 10 hours. The N1-mono- or N1,N1-di-substituted ethylene diamine starting materials may be prepared by saponifying the corresponding phthalimidoethyl derivatives obtained by reaction of 2-bromoethyl-phthalimide with the appropriate primary or secondary amine. Pharmaceutical compositions for oral or parenteral administration having hypotensive activity comprise the above guanidine compounds together with a pharmaceutically acceptable carrier. The compositions may be formulated as tablets, powders, pills, solutions or suspensions.
