abstract | A method of producing a rotavirus-like particle (RLP) in a plant, portion of a plant or plant cell comprising: ai) providing the plant, portion of the plant or plant cell comprising one or more nucleic acids comprising a A first nucleotide sequence that encodes a first rotavirus protein, a second nucleotide sequence that encodes a second rotavirus protein, a third nucleotide sequence that encodes a third rotavirus protein, and a fourth nucleotide sequence that encodes a fourth rotavirus protein. rotavirus, where the first, second, third, and fourth nucleotide sequences are operably linked to one or more active regulatory regions in the plant, portion of the plant, or plant cell; the first nucleotide sequence encodes the rotavirus NSP4 protein, the second nucleotide sequence encodes the rotavirus VP6 structural protein, the third nucleotide sequence encodes the rotavirus VP2 structural protein, and the fourth nucleotide sequence encodes the rotavirus VP7 structural protein ; or a-ii) introducing into the plant, portion of the plant or plant cell by transforming the one or more nucleic acids comprising the first nucleotide sequence encoding the first rotavirus protein, the second nucleotide sequence encoding the second rotavirus protein, the third nucleotide sequence encoding the third rotavirus protein, and the fourth nucleotide sequence encoding the fourth rotavirus protein, where the first, second, third, and fourth nucleotide sequences are operably linked to one or more active regulatory regions in the plant, portion of the plant or plant cell; and the first nucleotide sequence encodes the rotavirus NSP4 protein, the second nucleotide sequence encodes the rotavirus VP6 structural protein, the third nucleotide sequence encodes the rotavirus VP2 structural protein, and the fourth nucleotide sequence encodes the rotavirus structural protein VP7; eb) incubating the plant, portion of the plant or plant cell under conditions that allow the expression of one or more nucleic acids, so that each of NSP4, VP6, VP2 and VP7 are expressed, thus producing the RLP, the RLP comprises rotavirus proteins, rotavirus proteins consist of rotavirus structural proteins; and optionally c) harvesting the plant, plant portion or plant cell, and d) purifying the RLPs from the plant, plant portion or plant cell, wherein the RLPs have a size ranging from 70-100 nm. |
