abstract | An in vitro method for testing antigen-specific T cell activation, comprising the steps of: a. Disposing of a phagocytable particle, having a candidate antigen polypeptide intimately associated therewith, wherein the particle with the associated polypeptide has been subjected to a denaturation wash that results in a low enough endotoxin level not to interfere with subsequent steps; b. Arrange a viable antigen presenting cell; c. Contacting the washed particle with the antigen-presenting cell in vitro under conditions that allow phagocytosis of the particle by the antigen-presenting cell; d. Arranging a sample of T cells to be tested comprising viable T cells; and. Contacting the T-cell sample with the antigen-presenting cell that has contacted the particle in vitro under conditions that allow specific activation of T-cells in response to an antigen presented by an antigen-presenting cell; and f. Determine the degree of T cell activation in the T cell sample. |
