abstract | A method of increasing the sensitivity of cell-based detection of a botulinum toxin, comprising: (i) providing a transfected cell that produces a construct comprising; (a) a first end comprising an indicator-containing portion, wherein the indicator-containing portion exhibits a signal; and, (b) a cleavage site that interacts with botulinum toxin in a manner that produces cleavage of the reporter-containing portion of a remnant of the construct; (ii) exposing the transfected cell to botulinum toxin to a first toxin exposure temperature; iii) transitioning the temperature of the transfected cell from the first toxin exposure temperature to a second toxin exposure temperature; wherein the first toxin exposure temperature is lower than the second toxin exposure temperature; or wherein the first toxin exposure temperature is greater than the second toxin exposure temperature; and (iii) obtaining the signal from the portion containing the reporter, wherein the sensitivity of the response of the transfected cell to botulinum toxin increases in relation to the sensitivity of the transfected cell to botulinum toxin when kept at 37 ° C throughout exposure to botulinum toxin; where the first toxin exposure temperature is 37 ° C and the second toxin exposure temperature is up to 41 ° C, or where the second toxin exposure temperature is 37 ° C and the first Exposure temperature to toxin is up to 41 ° C. |
