abstract A method for detecting a nucleic acid methylation state in an automated system comprising an extraction chamber comprising an extraction membrane, said method comprising the steps of: - introducing a nucleic acid source into the automated system, adding bisulfite thereto and immediately followed by heating the nucleic acid source to produce a lysate comprising the nucleic acid and deamine an unmethylated cytosine base present in said nucleic acid; - adding binding buffer to the lysate to produce a lysate and binding buffer solution; - binding the nucleic acid to the extraction membrane and removing the bisulfite by transporting the lysate solution and binding buffer over the extraction membrane; - add desulfonation buffer to the extraction chamber and transport the desulfonation buffer over the extraction membrane to desulfonate the deaminated base in a previous step in the nucleic acid and convert said base to a uracil base, - elute the converted nucleic acid of the extraction membrane; and analyzing the converted nucleic acid.
