abstract A method of modifying a nucleic acid, comprising: (a) contacting a single-stranded nucleic acid with a terminal transferase activity and a mixture of two or more different nucleotides under conditions in which the nucleotides in the mixture add sequentially and randomly to the 3' end of the nucleic acid by terminal transferase activity, thereby adding a heteropolynucleotide comprising nucleotides in the mixture to the 3' end of the nucleic acid and generating a first modified nucleic acid; (b) contacting the modified first nucleic acid with a terminal transferase activity and a composition comprising single nucleotide monomers under conditions where the monomers add to the 3' end of the modified first nucleic acid via the transferase activity terminal, thereby adding a homopolynucleotide comprising the monomers to the 3' end of the first modified nucleic acid and generating a second modified nucleic acid comprising, 5' to 3', the heteropolynucleotide and homopolynucleotide; and (c) contacting the second modified nucleic acid with a second single-stranded nucleic acid comprising a sequence that is complementary to, or substantially complementary to, the homopolynucleotide of the second modified nucleic acid and a ligase activity, under conditions where the 3' end of the second modified nucleic acid and the 5' end of the second single-stranded nucleic acid are ligated, thereby generating a ligated nucleic acid, wherein the 5' end of the second single-stranded nucleic acid is phosphorylated or adenylated.
