abstract | A method comprising: receiving a fluorescence emission spectrum generated by a protein sample at a first excitation wavelength, said protein sample being configured to fluoresce depending on its conformational state; evaluating, from said fluorescence emission spectrum, a characteristic indication of a fluorescence response of said protein sample at said first excitation wavelength; repeating said receiving and evaluating steps with respect to a plurality of fluorescence emission spectra, each fluorescence spectrum generated by said protein sample at an excitation wavelength different from said first excitation wavelength; generating a non-linear relationship between the excitation wavelength and the characteristic indication of the fluorescence response of said protein sample, said non-linear relationship comprising an indication of at least one characteristic of said conformational state of said protein sample, based on in correlating said excitation wavelengths and associated evaluated indications characteristic of said fluorescence response of said protein sample; and determining a characterization of said conformational state of said protein sample based on the curvature of said generated non-linear relationship. |
