abstract A method of measuring amplification of a target nucleic acid molecule using a nucleic acid probe, the method comprising: amplifying the target nucleic acid molecule in the presence of a nucleic acid probe, wherein the nucleic acid probe hybridizes to the target nucleic acid molecule and has a probe:template melting temperature less than the denaturation temperature, annealing temperature, and extension temperature used in a polymerase chain reaction (PCR) amplification cycle, in wherein said method quantitatively measures the initial amount of said target nucleic acid by competitive quantitative PCR or real-time quantitative PCR.
