abstract | An RNA replicon comprising a modified alphavirus 5 'replication recognition sequence, wherein the modified 5' replication recognition sequence is characterized in that all initiation codons have been removed from conserved sequence element 2 (CSE 2) of the native alphavirus 5 'replication recognition sequence and wherein the modified 5' replication recognition sequence comprises one or more additional nucleotide changes that compensate for nucleotide pairing alterations within one or more introduced hairpins (SL) by the elimination of the initiation codons, wherein said CSE 2 spans from SL2 to SL4 and comprises the native initiation codon that encodes the first amino acid residues of the alphavirus nsP1 non-structural protein, and wherein the recognition sequence of Modified 5 'replication is characterized by a predicted secondary structure that is equivalent to the structure predicted secondary of the 5 'replication recognition sequence of alphaviral genomic RNA. |
