abstract | A method of assaying fetal extravillous trophoblast cell RNA, comprising: providing a maternal endocervical sample obtained from a pregnant subject; fix fetal extravillous trophoblast cells by treatment with an aldehyde fixative which produces aldehyde-fixed fetal extravillous trophoblast cells; removing fetal extravillous trophoblast cells from the maternal endocervical sample, producing isolated aldehyde-fixed fetal extravillous trophoblast cells; wash isolated aldehyde-fixed fetal extravillous trophoblast cells or wash a lysate of isolated aldehyde-fixed fetal extravillous trophoblast cells to promote removal of aldehyde fixative-introduced crosslinks, producing washed fetal extravillous trophoblast cells or a lysate washed; extracting fetal extravillous trophoblast cell RNA from washed fetal extravillous trophoblast cells or the washed lysate; and assaying fetal extravillous trophoblast cellular RNA for one or more characteristics of fetal extravillous trophoblast cellular RNA; where the treatment with the aldehyde fixative is carried out before the removal of the fetal extravillous trophoblast cells from the maternal endocervical sample. |
