abstract | A nucleic acid construct comprising the following structure: A-X-B in which X is a nucleic acid sequence encoding a cleavage site; and A and B are nucleic acid sequences encoding a first and a second chimeric antigen receptor (CAR), each CAR comprising: (i) an antigen-binding domain; (ii) a spacer (iii) a transmembrane domain; and (iv) an endodomain in which the antigen-binding domains of the first and second CAR bind to different antigens, in which the spacer of the first CAR is of a different size than the spacer of the second CAR so that when the first CAR and the second CAR bind to their respective target antigens on the cell surface, the first CAR and the second CAR are spatially separated on the cell membrane; wherein one of the first or second CAR is an activating CAR comprising an activating endodomain and the other CAR is an inhibitory CAR comprising an inhibitory endodomain in the absence of ligation, inhibitory CAR that inhibits the activation of T lymphocytes by the activating CAR in the absence of ligation by the inhibitory CAR, but does not significantly inhibit the activation of T lymphocytes by the activating CAR when the inhibitory CAR is bound; and wherein: (a) the first and / or second CAR comprises an intracellular retention signal; and / or (b) the signal peptide of the first or second CAR comprises one or more mutations such that it has fewer hydrophobic amino acids. |
