abstract A method of predictably inserting a single nucleotide into a target region at a target site in DNA after Cas9-mediated cleavage, the method comprising: selecting a gene comprising the target region to be modified; designing a guide polynucleotide to target a selected 20 nt protospacer in the target region; producing a double-stranded break at the target site in the target region using a Cas9 protein and the guide polynucleotide complementary to the protospacer in the target region such that the nucleotide at position 17 of the protospacer corresponds to an A or a T; wherein (i) for the insertion of an A at the double-stranded break at the target site, position 17 of the protospacer is A, or (ii) for the insertion of a T at the double-stranded break at the site target, position 17 of the protospacer is T; thus predictably inserting a single nucleotide into the target site after Cas9-mediated cleavage; wherein the method does not modify the genetic identity of the germ line of a human; wherein the method is not a method for treating the human or animal body by therapy.
