abstract | A composition for use in a method of treating a bleeding disorder in a mammal, wherein the composition comprises an adeno-associated virus (AAV) vector comprising a VP1, VP2 and VP3 sequence of AAV-Rh74 set forth in SEQ ID NO. : 1, 3 and 4, respectively, as shown in Figure 3, and said vector further comprises a heterologous polynucleotide sequence comprising a gene encoding a blood coagulation (clot-forming) factor, wherein the polynucleotide sequence heterologous is operably linked to an expression control element that confers transcription of said heterologous polynucleotide sequence, wherein the expression control element is a promoter active in the liver, and wherein the method comprises administering the adeno-associated virus (AAV) vector to said mammal, whereby the heterologous polynucleotide sequence is administered or transferred to the mammal. |
