abstract Selective isolation procedure of microorganisms of interest within a liquid biological sample that comprises or is capable of comprising, especially: * microorganisms of interest whose cell membrane or capsid does not contain cholesterol, and * non-target elements, that is: - non-target cells whose cell membrane contains cholesterol, and - optionally, cholesterol-containing envelope viruses, and - optionally cholesterol-containing mycoplasmas, and - optionally residues of microorganisms of interest and / or non-target cells, comprising the procedure involves the following steps: a) contact the liquid biological sample with a saponin formulation, in order to destabilize the cholesterol-containing cell membranes or cholesterol-containing viral envelopes or cholesterol-containing microplasm membranes, b) perform an osmotic shock of non-target cells in order to specifically lyse them, c) aƱ add a solution of at least one enzyme suitable to lyse the free nucleic acids, DNA and / or RNA, from the non-target elements lysed in solution in the sample, which allow to selectively obtain the microorganisms of interest, d) add an agent of precipitation of the microorganisms of interest not lysed in solution in the sample, said agent allowing the adhesion of the residue that contains these microorganisms of interest and choosing between polyethylene glycol (PEG), glycogen, nucleic acids or a mixture of these precipitation agents .
