abstract |
A method of producing recombinant AAV viral particles in a suspension-growing BHK cell, comprising: co-infecting a suspension-growing BHK cell with (i) a first replication-deficient, recombinant herpes simplex virus comprising an acid nucleic coding for an AAV rep gene and an AAV cap gene each operably linked to a promoter; and (ii) a second replication-deficient recombinant herpes simplex virus comprising a nucleic acid encoding a gene of interest operably linked to a promoter, said nucleic acid being flanked by the inverted terminal repeat sequences of AAV, wherein the cells are infected with the first rHSV and the second rHSV in a ratio of 1: 2 to 6: 1, for a combined MDI of between 3 and 14, thus producing recombinant AAV viral particles in a BHK cell, in which the number of viral particles produced is equal to or greater than the number of viral particles grown in an equal number of BHK cells under adherent conditions. |
