abstract | A method, comprising the steps of: a) introducing a culture medium, a lytic agent, and a cellular debris-cleaving enzyme into a blood sample, comprising erythrocytes and at least one living microbial cell, wherein the introduction of the lytic agent and cellular debris cleavage enzyme in the blood sample degrades erythrocytes to produce a concentration of free heme in the blood sample, which can reduce the growth of at least one live microbial cell, wherein the blood sample blood is a direct blood culture from the patient or a positive blood culture; b) introducing a heme polymerase to the blood sample; c) incubating the blood sample with the lytic agent and the cellular debris cleaving enzyme and heme polymerase for a period of time to produce a digested blood sample; d) causing the growth of the at least one live microbial cell for the period of time to produce a threshold number of live microbial cells necessary for identification; and e) determining the species and/or strain of at least one of the living microbial cells using automated digital microscopy. |
