abstract | A kit for the identification of an organism comprising: a) a set of primers for the amplification of a target nucleic acid sequence comprising a variable nucleic acid sequence, wherein the variable nucleic acid sequence varies among a group of organisms and is flanked by sequences that are at least relatively conserved among members of a group of organisms, where the primers in the primer set hybridize to the flanking sequences, and b) multiple detectably distinguishable probe sets, where the probes from the probe sets hybridize adjacently to each nucleotide of the variable nucleic acid sequence, and wherein each probe set comprises (i) a probe labeled with a non-fluorescent quencher, and (ii) a probe labeled with a fluorescent moiety where the probe labeled with a fluorescent moiety for the target sequence emits a signal over background if it hybridizes to the target sequence target nucleic acid in the absence of the non-fluorescent quencher labeled probe, and wherein, if both probes of a probe set hybridize to the target nucleic acid sequence, the non-fluorescent quencher quenches the signal from the fluorescent moiety. |
