abstract | A method of expressing two or more nucleic acid sequences of interest in eukaryotic cells, in cells or tissues of a eukaryotic organism, or in a eukaryotic organism excluding humans, comprising: (a) providing eukaryotic cells, tissue from a eukaryotic organism or a eukaryotic organism excluding humans comprising two or more nucleic acid constructs of interest, each nucleic acid construct comprising a promoter and, downstream thereof, a nucleic acid sequence of interest to be expressed, wherein each promoter of said nucleic acid constructs of interest are selected from a library of multiple promoters each comprising, in the 5 'to 3' direction, (i) an effector binding site comprising at least 10 contiguous bases, wherein the sequences base of the effector binding sites of multiple promoters have a sequence identity of at least 7 5%, (ii) a TATA box, and (iii) a downstream sequence segment comprising at least 20 contiguous bases, where the downstream sequence segments of at least two nucleic acid constructs of interest differ, and ( b) providing the eukaryotic cells, the tissue of a eukaryotic organism or a eukaryotic organism of the previous stage with an effector capable of binding to the effector binding sites of the promoters and activating the transcription of said promoters. |
