A method for producing a heterodimeric molecule comprising a CH3 domain from a single cell, wherein said molecule comprises two human IgG CH3 domains that are capable of forming an interface, said method comprising providing in said cell a. A first nucleic acid molecule encoding a first polypeptide chain comprising a CH3 domain of human IgG, b. A second nucleic acid molecule encoding a second polypeptide chain comprising a CH3 domain of human IgG, wherein said first polypeptide chain comprising a CH3 domain comprises a lysine residue (K) at amino acid position 366 according to the EU numbering system and wherein the second polypeptide chain comprising a CH3 domain comprises an aspartic acid residue (D) at amino acid position 351 according to the EU numbering system, and said method further comprises the step of culturing said host cell and allowing the expression of said two nucleic acid molecules and the collection of said heterodimeric molecule comprising a CH3 domain of the culture. |
