abstract A method for producing a therapeutic protein in a mammalian cell that endogenously expresses a cognate receptor of said recombinant therapeutic protein and where the binding of said therapeutic protein to said cognate receptor results in a low titre of the therapeutic protein, comprising the method with a mammalian cell that is deficient in the expression of the cognate receptor of said therapeutic protein and being transformed with an expression vector comprising a nucleic acid molecule encoding the therapeutic protein: a. Cultivate said cell under conditions that allow the expression of the therapeutic protein; and b. Collect the therapeutic protein from the mammalian cell cultured in step a, where said mammalian cell produces the therapeutic protein in at least a factor 1.5 more than the same cell in which the expression of the cognate receptor has not been modified from this way, where the therapeutic protein is the protein of the insulin growth factor 1 (IGF-1) or a variant thereof whose cognate receptor is the IGF-1R receptor; and where said mammalian cell is deficient in the expression of a functional insulin growth factor 1 (IGF-1R) receptor, the cognate receptor of IGF-1 or the variant thereof.
