abstract | A composition of two or more manipulated nucleic acid sequences that form a scaffold ("NASC"), the NASC composition comprising: a first manipulated nucleic acid component ("NASC-PC1") comprising, in a 5 'a direction 3 ', a spacer element 1 comprising a nucleic acid target binding sequence 1, a repeat element 1 comprising a repeat nucleic acid sequence 1, and a nucleic acid binding protein binding element 1 comprising a double-stranded nucleic acid binding protein sequence 1, in which the spacer element 1 is covalently connected with the repeating element 1, and the repetition element 1 is covalently connected with the binding element 1 to the nucleic acid binding protein; and a second manipulated nucleic acid component ("NASC-PC2") comprising, in a 5 'to 3' direction, a spacer element 2 comprising a nucleic acid target binding sequence 2, a repeating element 2 comprising a 1C sequence of repeat nucleic acids, and a nucleic acid binding protein 2 comprising a sequence 2 of double chain nucleic acid binding protein, in which the spacer element 2 is covalently connected with the repeating element 2, and the repeating element 2 is covalently connected with the nucleic acid binding protein binding element 2; in which there is a connection between the repeat nucleic acid sequence 1 and the repeat nucleic acid sequence 1C through base pairs with hydrogen bonds, the connection forms the NASC composition, and the NASC composition is able to bind to a first CRISPR-Cas9 Class 2 Type II protein and a second CRISPR-Cas9 Class 2 Type II protein. |
