A non-therapeutic method for inducing a mammalian cell to produce a recombinant protein, the method comprising contacting said mammalian cell with an in vitro synthesized mRNA comprising an open reading frame encoding said recombinant protein; wherein the mRNA synthesized in vitro comprises Ψ or m1Ψ (1-methylpseudouridine); and wherein the amount of IL-12, IFN-α, TNF-α, RANTES, MIP-1α, MIP-1ß, IL-6, IFN-β or IL-8 secreted in response to said contact is less than respective amount of IL-12, IFN-α, TNF-α, RANTES, MIP-1α, MIP-1ß, IL-6, IFN-β or IL-8 secreted in response to contact of said mammalian cells with a homologous mRNA unmodified that does not comprise Ψ or m1Ψ (1-methylpseudouridine). |
