abstract | An autonomous apparatus for isolating nucleic acid from at least one unprocessed sample, apparatus for use with an instrument, said apparatus consisting of a macro-fluid component, a microfluidic component, at least one pneumatic transmission line and two inputs, a first inlet. to receive the at least one unprocessed sample from a collection device and a second input to establish an interface in the transmission mechanism in said instrument: (i) said macro-fluid component comprising: at least one camera to receive said at least one sample unprocessed from a collection device; at least two storage tanks of pre-loaded lysis reagents formed within the macro-fluid component; a storage tank of pre-washed washed reagents within the macro-fluid component; and a storage tank of preloaded elution reagents within the macro-fluid component; (ii) a microfluidic component in communication with said macro-fluidic component through at least one microfluidic element, said microfluidic component further comprising at least one nucleic acid purification matrix; and (iii) at least one pneumatic transmission line for connection to a transmission mechanism in said instrument for transmitting said first and second lysis reagents, said washing reagent and said elution reagent sequentially through said microfluidic element and said nucleic acid purification matrix, in which the only inputs to said apparatus are by said chamber and said at least one transmission line and, additionally, where all the reagents used to isolate nucleic acid from the at least one unprocessed sample they are contained inside the device in preloaded cameras. |
