A method of characterizing a target polynucleotide, the method comprising: (a) applying a potential difference through a pore in contact with a Hel308 helicase and the target polynucleotide; (b) measure at least two signals of different magnitudes per nucleotide of the target polynucleotide that moves through the pore during a complete translocation cycle of Hel308 helicase; and (c) characterize the target polynucleotide using at least two signals of different magnitudes per nucleotide measured in (b). |
