abstract A recombinant single-chain meganuclease comprising: (a) a first LAGLIDADG subunit comprising a polypeptide sequence having at least 85% sequence identity with residues 9-151 of a wild-type I-CreI meganuclease of SEQ ID NO: 1 and which has a first recognition semisite; and (b) a second LAGLIDADG subunit comprising a polypeptide sequence having at least 85% sequence identity with residues 9-151 of a wild-type I-CreI meganuclease of SEQ ID NO: 1 and having a second recognition semisite; wherein said first and second LAGLIDADG subunits are covalently linked by a polypeptide linker; wherein said polypeptide linker comprises 23-56 residues; wherein said first subunit is covalently linked to said polypeptide linker in a moiety corresponding to a position selected from the group consisting of positions 142-163 of SEQ ID NO: 1; wherein said second subunit is covalently linked to said polypeptide linker in a moiety corresponding to a position selected from the group consisting of positions 1-9 of SEQ ID NO: 1; and wherein said first and second LAGLIDADG subunits are able to act together to recognize and cleave a non-palindromic DNA sequence that is a hybrid of said first recognition semisite and said second recognition semisite.
