Method for synthesizing an RNA molecule of given sequence, comprising the following steps: a) determining the fraction (1) for each of the four nucleotides G, A, C and U of said RNA molecule, and b) synthesizing said molecule RNA by in vitro transcription in an optimized sequence reaction mixture, wherein said optimized sequence reaction mixture comprises the four ribonucleoside triphosphate (NTP) GTP, ATP, CTP and UTP, where the fraction (2) of each of the four ribonucleosides triphosphate in the optimized sequence reaction mixture corresponds to the fraction (1) of the respective nucleotide in said RNA molecule, a buffer, a DNA template and an RNA10 polymerase. |
