abstract | The use of an RNAi agent to improve RNA silencing of a target mRNA in a non-therapeutic method of RNA silencing performed in a cell that has an RNAi path; wherein the RNAi agent has at least one terminal nucleotide substituted with a nucleotide that forms an oscillating base pair G: U with the corresponding nucleotide in the target mRNA, in which the terminal nucleotide is within 5 or less end nucleotides 3 'of the RNAi agent; wherein the RNAi agent has at least one terminal nucleotide substituted with a nucleotide that forms a pair of oscillating bases G: U with the corresponding nucleotide in the target mRNA, in which the terminal nucleotide is within 5 or less nucleotides of the 5 'end of the RNAi agent; and wherein at least two terminal nucleotides are substituted, in which the two substituted terminal nucleotides are at the 3 'end of the RNAi agent, wherein the method comprises contacting the cell with the RNAi agent under conditions such that silencing be improved. |
