abstract | In vitro procedure for determining the binding of a competitor antibody with an Fc receptor expressed in cell membranes or intact cells present in a measurement medium, by competition with a reference antibody, comprising the following steps: (i) direct labeling of said Fc receptor by the first member of a pair of FRET pairs, or by introducing intact cells or cell membranes into the medium whose Fc receptors have previously been directly labeled by the first member of a pair of pairs of FRET; (ii) addition in the measuring medium of the competitor antibody; (iii) addition in the measuring medium of a reference antibody, directly labeled by the second member of said pair of FRET pairs; (iv) measurement of the FRET signal, being a decrease in the signal measured in the presence of the competitor antibody with respect to the measurement in its absence representative of the binding of this antibody to the Fc receptor; characterized in that the Fc receptor is directly labeled by a suicide enzyme, namely, that it is expressed in the form of a fusion protein comprising the Fc receptor and the suicide enzyme, and that the labeling is performed by addition to the medium of substrate measurement of the enzyme, conjugated to one of the FRET pairs. |
