abstract |
An affinity chromatography matrix comprising a solid support with an affinity chromatography ligand that comprises either one or more B domains of Staphylococcus aureus Protein A (SpA) with the amino acid sequence shown in SEQ ID NO .: 3 or encoded by the nucleotide sequence shown in SEQ ID NO .: 9, or one or more Z domains of SpA with the amino acid sequence shown in SEQ ID NO .: 6 or encoded by the nucleotide sequence shown in SEQ ID NO .: 12, where at least one of the one or more domains is mutated to delete three, four or five consecutive amino acids from the N-terminus, starting at position 1 or position 2 of SEQ ID NO: 3 or SEQ ID NO.:6; wherein the ligand exhibits reduced fragmentation with respect to a wild-type equivalent, after exposure to 0.5M NaOH for at least 5 hours. |
