abstract | A method of sequential isolation and purification of proteins comprising (i) providing a plasma sample, (ii) providing ligands that bind, each of them, specifically to a target protein of the biological sample, where each of the ligands is optionally attached to a support to form ligand-support complexes, (iii) contacting ligands or ligand-support complexes, sequentially and in a predetermined order, with the plasma sample to allow each ligand or ligand-support complex to selectively the target protein of the plasma sample, where the predetermined order of contacting ligands or ligand-support complexes with the plasma sample results in plasminogen binding before IgG and IgG before albumin, and where, before of said contacting, the biological sample is not processed by a preconditioning stage selected from the group consisting of precipitation with alc ohol, cryoprecipitation, elimination of lipids and / or lipid proteins, precipitation of euglobulin, or a combination thereof, (iv) elute the bound protein bound to each of the ligands or ligand-support complexes, and (v) isolate the target protein sequentially from the biological sample. |
