abstract A method comprising (a) contacting (1) a fusion protein comprising a polypeptide and an N-fragment of divided intein comprising a sequence selected from the group consisting of SEQ ID NO: 1-19, or a variant of the same, in which the variant comprises a sequence selected from the group consisting of SEQ ID NO: 21-38 and 761 and (2) a fragment C of divided intein comprising a sequence selected from the group consisting of SEQ ID NO: 57-74, in which the contact is made under conditions that allow for the attachment of the N fragment of the divided intein to the C fragment of the divided intein to form an intein intermediate; and (b) contacting the intein intermediate with a nucleophile to form a protein and nucleophile conjugate.
