abstract | An in vitro method for obtaining a population of stromal progenitor cells from a first selected amount of adipose tissue containing fat cells, the method comprising the steps of: - digesting said first amount with a second amount of enzyme buffer of the type used in the dissociation of organic and biological tissues to obtain a population of free cells; - sow this population of free cells in culture media necessary for its expansion; characterized in that said adipose tissue is white adipose tissue of subcutaneous origin and said first quantity is between 0.004 ml and 0.036 ml and that second quantity is between 0.9 ml and 1.1 ml, and because said second quantity comprises solvent and soluble media that comprise: Dulbecco-modified Eagle's medium (DMEM) penicillin (1U / ml) and streptomycin (1 mg / ml) sodium pyruvate (1 mM) non-essential amino acids comprising: L-alanine (0.89 mg / dl); L-asparagine H20 (1.5 mg / dl); L-aspartic acid (1.33 mg / dl); L-glutamic acid (1.47 mg / dl); glycine (0.75 mg / dl); L-proline (1.15 mg / dl); L-serine (1.05 mg / dl) and wherein said soluble media comprise: - mixture of collagenase class I and II; and because - said mixture of class I and II collagenase has an enzymatic activity between 0.1 U / mg and 5.5 U / mg of lyophilized product, which has an enzymatic concentration between 1.74 and 1.78 U / ml of said solvent medium and wherein said soluble media comprise: clostripain at a concentration between 0 U / mg and 7.6 U / mg of lyophilized product; - proteases at a concentration between 0.05 U / mg and 164 U / mg of lyophilized product; - trypsin at a concentration between 0.23 U / mg and 2.3 U / mg of lyophilized product in a form selected from individual agents or in combination. |
