abstract | A cryopreservation method of an equivalent of organotypically cultured skin to maintain a viable tissue comprising: a) treating an equivalent of organotypically grown skin in a single stage cryoprotective solution, wherein said equivalent of organotypically cultured skin comprises stratified squamous epithelium on a dermal layer comprising fibroblasts and wherein said cryoprotectant is provided in a solution comprising from 21% to 70%, preferably from 21% to 45% or from 37.5% to 62.5%, most preferably of the 25% to 40% or 42.5 to 57.5% of said volume solution and said cryoprotectant is glycerol; b) separating the equivalent of organotypically treated cultured skin from the excess of cryoprotective solution and packaging said equivalent of organotypically treated cultured skin in the absence of additional cryoprotectant to provide an equivalent of packaged skin, in which said package preferably further comprises enclosing said skin equivalent cryopreserved in a sterile bag and enclose said sterile bag in a second bag; and c) freezing said equivalent of organotypically packaged cultured skin to provide an equivalent of cryopreserved skin. |
