abstract | A dual construction system for expressing the coding sequence of a gene of interest in a host cell, and said coding sequence consists of a portion of the 5 'end and a portion of the 3' end, and said dual construction system comprises: a) a first plasmid comprising, in the 5'-3 'direction: - a 5' (5'-ITR) inverted terminal repeat sequence of AAV; - a promoter sequence; - the 5 'end portion of said coding sequence, and said 5' end portion is operably linked to said promoter and is under its control; - a nucleic acid sequence of a donor signal breaking; and - a 3 '(3'-ITR) inverted terminal repeat sequence of AAV; and b) a second plasmid comprising, in the 5'-3 'direction: - a 5' (5'-ITR) inverted terminal repeat sequence of AAV; - a nucleic acid sequence of a breaker acceptor signal; - the 3 'end of said coding sequence; - a polyadenylation signal nucleic acid sequence; and - a 3 '(3'-ITR) inverted terminal repeat sequence of AAV, wherein said first plasmid further comprises a nucleic acid sequence of a recombinogenic region at the 5' position of the AAV 3'-ITR of said first plasmid and at the 3 'position of the nucleic acid sequence of the donor signal breaking, and wherein said second plasmid further comprises the nucleic acid sequence of the recombinogenic region at the 3' position of the 5'-ITR of AAV said second plasmid and the 5 'position of the nucleic acid sequence of the acceptor break signal, wherein the recombinogenic region is a recombinogenic region of phage F1 consisting of the sequence: GGGATTTTGCCGATTTCGGCCTATTGGTTAAAAAATGAGCTGATTTAACAAAAATTTAACGCGAATTTTAACAAAA T (SEQ ID NO: 3), or one of its fragments that maintains the recombinogenic property of SEQ ID NO: 3, wherein, after the introduction of said first plasmid and said second plasmid into cell h ospedante, said coding sequence is reconstituted by means of the donor signals of rupture and acceptor of rupture. |
