abstract | An isolated modified active Factor Xa (FXa) polypeptide comprising an amino acid substitution at a position corresponding to position 196 and at a position corresponding to position 332 in an unmodified FXa polypeptide with reference to the positions of amino acids shown in SEQ ID NO: 134, wherein: the corresponding amino acid positions are determined by alignment with SEQ ID NO: 134; the substitution at position 196 is with a neutral polar amino acid residue that is serine (S) or threonine (T); the substitution at position 332 is Alanine (A), Aspartate (D), Glutamate (E), Serine (S) or Glycine (G); The unmodified FXa comprises a light chain consisting of the amino acid sequence shown as residues 1-139 of SEQ ID NO: 134, and a heavy chain consisting of the amino acid sequence shown as residues 195 -448 of SEQ ID NO: 134, wherein: the modified FXa has at least a 50-fold increased Fa cofactor dependence compared to the unmodified FXa polypeptide that does not contain the amino acid substitution; and the modified FXa comprises two, three, four, five, six, seven, eight, nine, ten, eleven, twelve, thirteen, fourteen, fifteen, sixteen, seventeen, eighteen, nineteen, or twenty amino acid substitutions compared to the unmodified FXa polypeptide. |
