Diagnostic assay of thymidine kinase for gene therapy applications |
abstract |
A retroviral particle comprising an HSV-TK polynucleotide comprising a nuclear export sequence (NES), and an HSV-TK substrate bound to a radioactive indicator; for use in a procedure to identify a patient with lesions capable of benefiting from treatment with gene therapy, said procedure comprising: a) administering to the patient the retroviral particle comprising an HSV-TK polynucleotide comprising a nuclear export sequence (NES ); b) administer the HSV-TK substrate attached to a radioactive indicator to the patient; c) measure the relative amount and location of the radioactive signal present in the patient; and d) determine the location of the lesions in the patient, where patients who have (i) a radioactive signal location correlate with the lesions measured in stage (d), and (ii) radioactive signals above a certain threshold ; They identify themselves as capable of benefiting from gene therapy treatment. |
