titleOfInvention | Means and methods for the generation of mammalian producer cells for the production of recombinant proteins |
abstract | A method for the production of a protein of interest in a mammalian producer cell, comprising the following process steps: a) providing a mammalian producer cell, b) increasing the concentration of at least one miR selected from the group consisting of miR-15, miR-16 and miR-34 in the mammalian producer cell provided in step a) by transfecting the mammalian producer cell with at least one miR selected from the group consisting of miR-15, miR-16 and miR-34 or with at least one vector containing a nucleotide sequence encoding at least one miR under the control of at least one regulatory element that allows overexpression of at least one miR, wherein the at least one miR is selected from the group consisting of miR-15, miR-16 and miR34 such that a genetically modified mammalian producer cell for protein production is obtained, c) culturing the mod mammalian producer cell genetically engineered for protein production, under conditions suitable for the production of the protein of interest, and d) recovering the protein of interest produced in step c), where the mammalian-producing cell is a rodent cell, a hamster cell , a mouse cell or a human cell, a Y0 cell, a COS cell, an sp2 / 0 cell, a CHO cell, a CHO-K1 cell or a CHO-K1SV cell, a CHO DG44 cell, a CHO DUXB11 cell, a gene knockout CHO GS cell, a gene knockout CHO FUT8 G2 cell, an NS0 cell, a PERC6 cell, a HEK293 cell, an HT1080 cell, or a BHK cell. |
