abstract | A recombinant polynucleotide vector for integrating a target gene into a locus of the heavy chain of the chicken immunoglobulin gene, comprising: (a) a region of the nucleic acid sequence downstream of the chicken immunoglobulin VH gene that comprises a sequence homologous to the 5 'side of the start codon of endogenous genomic DNA encoding a chicken immunoglobulin VH gene; (b) a target gene comprising a rearranged chicken immunoglobulin VH-D-JH gene that has been isolated from a population of cells of the chicken bag of Fabrizio, wherein said VH-D-JH gene is rearranged accordingly so that the genes VH, D and JH are together; and (c) a region of the nucleic acid sequence downstream of the chicken immunoglobulin JH gene comprising a sequence homologous to the 3 'side of the splice site of endogenous genomic DNA encoding a JH immunoglobulin gene of chicken, in which the target gene, after being integrated into the chicken immunoglobulin heavy chain locus of a DT40 cell, is capable of undergoing either or both of (i) somatic hypermutation in a coding sequence of the VH region immunoglobulin, and (ii) gene conversion between a rearranged chicken immunoglobulin VH encoding nucleic acid sequence and a DT40 VH pseudogene nucleic acid sequence. |
