abstract | A method of detecting a target nucleic acid, the method comprising amplifying the target nucleic acid in the presence of a DNA polymerase having error-correcting activity, and an allele-specific reactive primer (ASRP) comprising i) a complementary nucleotide sequence to the target nucleic acid and ii) a nucleotide substituted with a 3'-deoxynucleotide, wherein an amplification product of the target nucleic acid is produced when the allele-specific reactive primer is complementary to the target nucleic acid, and when the allele-specific reactive primer of the allele is not complementary to the target nucleic acid, a nucleotide non-complementary to the target nucleic acid present at the 3' end of the primer is removed by the error-correcting activity of DNA polymerase, and an acid amplification product is not produced. target nucleic due to the 3'-deoxynucleotide remaining at the 3' end of the primer. |
