abstract | A method of detecting in a sample a methicillin-resistant Staphylococcus aureus (MRSA) having an insertion of a SCCmec module within the chromosomal DNA of Staphylococcus aureus, the method comprising A) performing in a sample an amplification reaction to detect MRSA that can amplify only (1) a connection of an inserted SCCmec module and Staphylococcus aureus chromosomal DNA and (2) a region of mecA, and B) detect, within the amplification products, the presence or absence of each of the junctions and mecA, wherein if the sample contains MRSA, the presence of both junction and mecA is detected in the sample, wherein the procedure comprises the steps of: contacting, in a single container, the biological sample with a) a first set of oligonucleotides comprising (1) a first oligonucleotide having a nucleic acid sequence that specifically hybridizes to a connecting region at the end of an SCCmec module, and (2) a second oligonucleotide having a nucleic acid sequence that specifically hybridizes to a region of Staphylococcus aureus chromosomal DNA flanking said SCCmec module to form a first reaction product of the biological sample and the first and second oligonucleotides, and b) a second set of oligonucleotides comprising (3) a third oligonucleotide having a nucleotide sequence that specifically hybridizes to a first nucleic acid region of mecA and (4) a fourth oligonucleotide having a nucleotide sequence which specifically hybridizes to a second nucleic acid region of mecA to form a second reaction product of the biological sample and the third and fourth oligonucleotides wherein the contacting step further comprises performing a transcription-based amplification reaction such as NASBA or DNA-NASBA using oligonucleotide sets (a) and (b) as primers; and identifying the presence of MRSA by detecting both a first and a second reaction product, wherein the identifying step comprises detecting the presence or absence of an amplification product from both sets of oligonucleotides. |
