abstract |
A method of preparing a recombinant autonomous parvovirus characterized in that (i) HEK 293T cells are transfected with (a) a recombinant parvovirus vector obtained from the rat parvovirus H-1 (H-1) or the tiny mouse virus ( MVM) that lacks the genes of the virus VP1 and VP2 capsid proteins but that conserves the coding sequences of the nonstructural protein NS1 / 2, b) a vector that contains the genes that encode the VP1 capsid proteins and Parvovirus VP2, and (c) a helper vector obtained from adenovirus comprising the Ad5 adenoviral DNA sequences of the E2a gene, the E4 gene (orf6) and a VAI RNA gene; and (ii) the recombinant parvovirus lacking the VP1 and VP2 genes but retaining the NS1 / 2 coding sequences is isolated from HEK 293T cells or the medium after culturing the cells. |
