abstract | Method for generating a recombinant bunyavirus replicon particle, the method comprises: A) providing a eukaryotic cell with growth medium; B1) providing T7 polymerase in the eukaryotic cell, B2) providing bunyavirus Gn and Gc proteins, optionally in combination with one or more NSm proteins, collectively referred to as (NSm) GnGc, in the eukaryotic cell; B3) providing a vector in the eukaryotic cell that encodes a segment of the bunyavirus L genome, which is flanked by a T7 promoter and cDNA of a ribozyme sequence; B4) provide a vector in the eukaryotic cell that encodes at least part of a segment of the bunyavirus S genome comprising the N gene and the 3 'and 5' URTs, which are flanked by a T7 promoter and cDNA of a sequence of ribozymes; B5) with the proviso that, if a segment of the bunyavirus M genome is provided in the eukaryotic cell, a vector is provided in the cell that encodes a segment of the bunyavirus M genome of which the GnGc coding region (i) does not it is expressed because it is not transcribed or not translated, or (ii) encodes a non-active Gn and Gc protein, which is flanked by a T7 promoter and ribozyme cDNA; C) generating a recombinant bunyavirus replicon particle; D) isolating the recombinant bunyavirus replicon particle from the growth medium; wherein the sequence of steps B1, B2, B3, B4, B5 is random and all or part of these steps can be performed simultaneously. |
