abstract An expression cassette comprising a promoter, a polynucleotide sequence encoding a polypeptide, and an expression enhancer element in which the expression enhancer element comprises a genomic DNA sequence not translated downstream of a glyceraldehyde 3 promoter. - mammalian phosphate dehydrogenase (GAPDH), in which the polypeptide encoded by the polynucleotide sequence is not GAPDH, and in which the genomic DNA sequence downstream of the mammalian GAPDH promoter is initiated in a region spanning from the position of nucleotide of approximately +1 to the nucleotide position of approximately +7000, in which the nucleotide position is relative to the start of transcription of the GAPDH mRNA, and in which the length of the genomic DNA sequence not translated downstream of the mammalian GAPDH promoter is from about 200 to about 2800 nucleotides, and in which the expression cassette further comprises plus an untranslated genomic DNA sequence upstream of a mammalian GAPDH promoter, in which the untranslated genomic DNA sequence upstream of the mammalian GAPDH promoter is initiated in a region spanning from about the 5 'end of the GAPDH promoter from mammalian to the nucleotide position of approximately -3500, in which the nucleotide position is relative to the start of transcription of the GAPDH mRNA, and in which the length of the untranslated genomic DNA sequence upstream of the GAPDH promoter Mammalian is from about 200 to about 2800 nucleotides.
