titleOfInvention Procedure to evaluate absorption properties of low solubility compounds
abstract A method for evaluating the effect of excipients, pH and combinations thereof on the predicted absorption properties of compounds of low solubility, the method comprising the step of evaluating a change in a flow function for a combination of a compound of low solubility and an excipient, the flow function corresponding to particles or matter that pass a barrier per unit area and unit of time, in which the barrier is selected from the group consisting of human tissues, animal tissues, plant tissues, cell culture models , and artificial membranes, and wherein evaluating the change in flux function comprises the steps of: a. preparing a calibration donor solution at a predefined pH value comprising the compound; b. preparing a donor solution at a predefined pH value comprising the compound and the excipient; c. prepare a receptor solution at the predefined pH value free of the compound and the excipient; d. providing the calibration donor solution in a first donor chamber separated from a first receiving chamber by the barrier, providing the donor solution in a second donor chamber separated from a second receiving chamber by the barrier, and providing the receiving solution in the first and second receiving chamber; and. incubating said solutions for a predefined period of time; and f. measuring the response of the compound in the receptor solution of the first and second receptor chambers, wherein measuring the response of the compound in the receptor solution of the first and second receptor chambers comprises measuring spectroscopic properties of the receptor solution, leading repeatedly carrying out steps (a) through (f) at a plurality of predefined pH values, and determining a relationship between the respective measurement of the response of the compound in the receiving solution of the first receiving chamber and the corresponding measurement of the response of the compound in the receptor solution of the second receptor chamber.
