abstract | In situ hybridization method for determining the presence of a chromosomal aberration in chromosomal DNA, comprising: - providing at least one molecular probe that hybridizes with a nucleic acid sequence associated with chromosomal aberrations, - providing chromosomal DNA, - providing a composition hybridization comprising from 1% (v / v) to 95% (v / v) of at least one polar aprotic solvent having a cyclic base structure, and a hybridization solution, wherein the polar aprotic solvent is selected from the group consisting of: ** (See formula) ** where the hybridization solution comprises at least one additional component selected from the group consisting of: buffering agents, accelerating agents, chelating agents, salts, detergents and blocking agents, and with the proviso that the hybridization composition contains less than 10% formamide, - combining the molecular probe, chromosomal DNA and the hybrid composition Riding for at least a sufficient period of time to hybridize the molecular probe with the target, and - detect the at least one molecular probe, - and determine the presence of chromosomal aberration. |
