abstract | A method for the recombinant production of a complex comprising i) a fusion polypeptide comprising in the N-C end direction a peptide that elicits T-lymphocyte response, (β2 microglobulin and the α1, α2 and α3 extracellular domains of a molecule of MHC class I with a relative frequency of 1% or more, ii) a pair of disulfide-linked polypeptide chains derived from an antibody hinge region, and in which each disulfide-linked polypeptide chain comprises a CH2 domain and a CH3 domain of human origin, in which one CH3 domain comprises the T366W mutation and the other CH3 domain comprises the T366S, L368A and Y407V mutations (numbers according to the Kabat EU index); and iii) at least one pair of a variable domain of the antibody light chain and a variable domain of the antibody heavy chain in a eukaryotic cell, comprising the following steps: - culturing a eukaryotic cell comprising one or more nucleic acids encoding the complex, and - recovering the complex from the cell or culture medium, in which the complex comprises exactly a fusion polypeptide of a peptide that elicits T lymphocyte response, (ß2 microglobulin and α1, α2 extracellular domains and α3 of a MHC class I molecule, and in which the fusion polypeptide - covalently binds either the C-terminus or the N-terminus of one of the disulfide-linked polypeptide chains, or - covalently binds the N-terminus of an antibody variable domain that is the variable domain of the heavy or light chain complementary to that comprised in the first disulfide-linked polypeptide chain, or - covalently binds to the former C-segment of a constant antibody domain that is the constant domain of the heavy or light chain complementary to that comprised in the first disulfide-linked polypeptide chain. |
