abstract | A genetically modified rodent comprising in its germ line an immunoglobulin light chain locus comprising (i) a single rearranged gene region of human immunoglobulin light chain variable region comprising human VL and JL segment sequences and (ii) a immunoglobulin light chain constant region gene sequence in operative binding to the single human immunoglobulin light chain variable region rearranged gene sequence, in which the only human immunoglobulin light chain variable region rearranged gene sequence comprises at least one Genetically modified insertion or replacement of a histidine codon that is not encoded by corresponding human germline VL and JL gene sequences, in which the gene sequence of rearranged variable region human immunoglobulin light chain alone encodes an immunoglobulin chain that understand a domi variable light chain nio, in which the histidine codon is in the nucleotide sequence encoding a complementarity determining region (CDR) of the light chain variable domain, in which the light chain variable domain comprises at least one histidine residue at an amino acid position encoded by the at least one inserted or substituted histidine codon, and in which the immunoglobulin light chain encoded by the immunoglobulin light chain locus is capable of pairing with a plurality of heavy chains of immunoglobulin selected by the rodent, in which each of the plurality of immunoglobulin heavy chains specifically bind different epitopes when paired with the immunoglobulin light chain. |
